Scientists from Imperial College London used psilocybin from magic mushrooms and/or magic truffles on a small group of patients suffering from depression. These scientists examined the effects on the brain.

The opinion of these researchers, as well as ours, is that you should not initially turn to magic mushrooms or magic truffles, with or without guidance. First, look at what you can do regarding your gut flora (diet, probiotics, etc.), exercise, and mindset. Feel free to read our free blogs for more information. We can also be of service by advising you. This can be done through an intake interview, after which you will receive a personalized plan. We can best conduct a magic truffle trip session when the foundation is in order.

The head of the Department of Psychedelic Research at Imperial College London, Dr. Robin Carhart Harris, draws the following conclusion:

We have shown for the first time clear changes in brain activity in depressed people treated with psilocybin after failing to respond to conventional treatments. Several of our patients described feeling 'reset' after the treatment and often used computer analogies.
For example, one said he felt like his brain had been 'defragged' like a computer hard drive, and another said he felt 'rebooted.'‘ Psilocybin may be giving these individuals the temporary 'kick start' they need to break out of their depressive states and these imaging results do tentatively support a 'reset' analogy. Similar brain effects to these have been seen with electroconvulsive therapy.‘

Psilocybin, the active ingredient in magic mushrooms and truffles, offers unique healing for depression on multiple levels: neurochemical, psychological, and physiological. What makes this molecule so special?

Neurochemical reset via 5-HT2A

After conversion in the body, psilocin binds to 5-HT2A receptors, especially in the default mode network (DMN) — the brain network that is hyperactive in depression. This connection causes temporary disorganization of rigid thought patterns and stimulates new brain connections, creating space for behavioral change and recovery.

Increased neuroplasticity with BDNF and glutamate

Psilocybin increases the release of glutamate and stimulates the production of via the 5-HT2A receptor BDNF (Brain Derived Neurotrophic Factor). This creates a structural more flexibility in brain networks — a crucial factor in breaking free from depressive patterns.

Inflammation inhibition in the brain

Under the influence of psilocybin, inflammatory markers decrease as TNF-α, IL-6 and CRP, what the neuroinflammation reduces in depression. This occurs partly through inhibition of STATE3 and NF-κB signaling pathways. Less inflammation = better functioning of mood, memory, and motivation.

HPA axis normalization

The HPA axis (the stress axis) becomes dysregulated in many depressed people. Psilocybin helps calm this system, partly via the amygdala and hippocampus, which leads to less chronic stress and more resilience.

Psychological transformation

In addition to biochemistry, the experience itself — provided it is properly supervised — ensures deep emotional processing, self-insight and reinterpretation of traumas. This is reinforced by increased oxytocin and dopamine, which restores feelings of connectedness and motivation.

Epigenetic impact

Studies suggest that psilocybin also gene expression modifiesThere is increased expression of BDNF genes and possible suppression of inflammatory genes. This explains why one or two sessions sometimes result in improvement lasting for months.

With Triptherapie, this effect is utilized in combination with lifestyle improvements, neurochemical support via supplements, and intensive preparation. You can read more about this in the detailed explanation regarding depression treatment with psilocybin and the underlying neuroplasticity and psychedelics.
