What is a macrodose?

A macrodose refers to a higher dosage of psychedelics, such as psilocybin, LSD or other psychedelics, that is taken to experience significant effects. Unlike microdosing, which involves taking sub-threshold amounts that may not produce noticeable effects, macrodosing aims to exceed the threshold level, allowing the user to fully engage with the psychedelic experience. This approach is used in therapeutic settings to facilitate deep psychological insights and emotional healing.

The different levels of macrodosing, also referred to as trip levels, are as follows:
  1. Triplevel 1: This is the lowest dose at which negative feelings of a serotonin deficiency disappear. The communication between the hemispheres of the brain changes, causing music to sound deeper and colors to become brighter.
  2. Triplevel 2: At this dose, one experiences brighter perception of colors and visual effects, such as moving objects. 2-dimensional patterns can also be perceived with closed eyes.
  3. Triplevel 3: This level includes very clear hallucinations, where objects distort and time distorts, making everything seem to take place in the "now".
  4. Triplevel 4: At a high dose, there are strong hallucinations, such as objects merging into each other. The perception of the ego fades.
  5. Triplevel 5: This is the strongest trip, where there is a total loss of visual connection with reality and users merge with space or the universe.

Macrodosing of psychedelics refers to ingesting a high, consciousness-altering dose of substances such as psilocybin (magic mushrooms), LSD, or DMT. This contrasts with microdosing, where users take approximately 5-20% of a standard dose to achieve subtle effects without a full trip experience.

The effectiveness of Macrodosing

Macrodoses often cause profound psychological experiences such as:

  • Ego dissolution (loss of self-awareness)
  • Visual hallucinations
  • Emotional catharsis
  • Mystical experiences

Research shows that these intense experiences can bring about lasting positive changes in:

  • Depression and anxiety symptoms (up to 80% improvement in some studies)
  • Addictive behavior
  • Post-traumatic stress
  • Existential anxiety in terminal patients

Macrodoses fundamentally influence the brain:

  1. Increased connectivity between brain regions

  2. Inhibition of the default mode network (associated with self-reflection)

  3. Stimulation of neuroplasticity via 5-HT2A receptors

Aspect Macrodosing Microdosing
Therapeutic effect Significant and long-lasting (months) Subtle and fleeting
Cognitive impact Deep mindset change Slight improvement in mood
Scientific evidence Strong clinical evidence Mainly anecdotal
Risk profile Higher (set/setting crucial) Lower

Researchers explain the stronger effect of macrodosing by the combination of:

  1. Pharmacological action on serotonin receptors

  2. Psychological meaning-making during the trip

  3. Neuroplastic changes that enable behavioral adaptation
